体外培育牛黄制剂对血吸虫病家兔肝纤维化门静脉壁ERK_(1/2)表达的影响 被引量:3

摘要 目的:探讨血吸虫病家免肝纤维化门静脉高压时门静脉壁ERK1/2的表达及体外培育牛黄复方制剂(In-vitrocultivatedcalculusboviscompoundpreparation,ICCBco)对其影响。方法:血吸虫尾蚴皮肤敷贴法感染14只家兔构建肝纤维化模型。其中7只家兔于感染后40d开始给以ICCBco喂养。分别用Westernblot及免疫组织化学方法检测家兔门静脉壁ERK1/2及C-fos的表达。结果:血吸虫病家兔形成过和程中门静脉壁ERK1/2活性较健康家兔明显增高,ICCBco可以降低ERK1/2活性。血吸虫病组门静脉壁pERK1/2光密度值较ICCBco干预组和正常对照组高2~3倍(P<0.01)。C-fos的平均染色指数由模型组的10.3716±0.9375降低到治疗组的5.6921±0.7924,差异有显著性(P<0.01)。结论:提示ICCBco可降低ERK1/2/C-fos活性,并可能通过影响ERK1/2/C-fos信号途径来干预血吸虫性门脉高压性血管病变的形成。 Objective To investigate the effects of in-vitro cultivated calculus bovis compound preparation(ICCBco) on expression of ERK1/2 of portal vein with experimental liver fibrosis in rabbits with schistosomia japonicum. Methods the model of liver fibrosis, which was percutaneously induced by the infection ofcercaria of s. japonicum in rabbit, and then six of them started to be treated with ICCBco by breeding at 60 d after the infection 40 d. The expression of ERK1/2 of portal vein were determined by western blot and immunohistochemical examination, respectively. Results The expression of ERK1/2 increased in portal vein from rabbits with schistosomiasis, and reversed by ICCBco therapy. The expression of ERK1/2 was decreased significantly after the ICCBco therapy. Conclusions ERK1/2/c-fos signal transduction pathway may play an important role in the pathogenesis of portal vein with experimental liver fibrosis in rabbits with s. japonicum, and ICCBco can effectively decrease the the expression of ERK1/2 of portal vein in rabbit with schistosomiasis.
