切断左侧穹隆-海马伞模型大鼠在补充雌二醇及中药巴戟天和川芎汤干预后大脑皮质躯体感觉诱发电位的变化 被引量:1

摘要 目的:观察切断左侧穹隆-海马伞模型大鼠在补充雌二醇、灌服中药巴戟天和川芎汤剂、生理盐水等3种状态下大脑皮质躯体感觉诱发电位的变化。方法:实验于2002-09/2003-01在青岛大学医学院附属医院脑血管病研究所完成。29只Wistar大鼠随机分为4组:前3组在相同条件下行左侧穹隆-海马伞切断。①雌二醇组:8只,大鼠当日术后开始补充外源性苯甲酸雌二醇(1mg/kg),皮下注射,1次/周,共4次。②中药干预组:8只,由补肾益精中药巴戟天和活血行气中药川芎水煎浓缩至每毫升含生药1g的汤剂,每次2.2mL,于当日术后开始灌胃,1次/d,共30次。③生理盐水组:6只,等容量生理盐水替代雌二醇,其余用法同雌二醇组。④假手术组:7只,不用任何药物,动物除了不切断穹隆-海马伞外,其余处理同以上各组。所有动物均在用药第30天时观察大脑皮质躯体感觉诱发电位。结果:29只大鼠均进入结果分析。①中药干预组和生理盐水组左侧皮质躯体感觉诱发电位波幅显著小于假手术组犤(27.34±8.50),(19.25±20.75),(65.23±27.75)μV,P均<0.01犦,生理盐水组右侧皮质躯体感觉诱发电位波幅显著小于雌二醇组和中药干预组犤(21.94±18.21),(84.73±62.91),(42.85±7.52)ms,P均<0.05犦。②各组之间躯体感觉诱发电位潜伏期较接近(P>0.05)。结论:①大鼠行左侧穹窿海马伞切断后,左、右两侧皮质躯体感觉诱发电位波幅均降低,在中药干预状态下,大鼠左侧皮质躯体感觉诱发电位波幅显著小于假手术组,而与雌二醇组比较没有显著差异,说明巴戟天、川芎虽然难于逆转穹隆-海马伞切断造成的感知通路损伤,但可以减轻损伤。②中药治疗后右侧皮质躯体感觉诱发电位波幅与假手术组接近,可能与右侧穹隆-海马伞传导通路的完整性有关。③补充外源性雌二醇组左右两侧皮质躯体感觉诱发电位波幅与假手术组接近,说明雌二醇可以调节胆� AIM: To observe the ehange of somatosensory evoked potentials of eortex in rats with fimbria/fornix transection in different conditions such as estradiol, medicinal indianmullberry root and chuanxiong tang , normal saline and so on . METHODS: The experiment was conducted in the Institute of Cerebrovaseular Disease, Affiliated Hospital of Qingdan University Medical College from September 2002 to January 2003. Twenty-nine Wistar rats were randomly divided into four groups: Rats in the prior 3 groups were performed left-side fimbria-fornix transection.① Estradiol group: 8 rats. eete, genous estradinl benzoate(1 mg/kg) was injected suntaeutaneously intn the rats after operation immediately, once a week . totally 4 times.②Interventinn group with traditional Chinese medicine: 8 rats. 2.2 mL of Chinese medicine was stomached-perfused into the rats after operation immediately,once a day. totally 30 times. The Chinese medicine was condensed from qi-replenishing and essence-nourishing medicinal indianmultberry root and circulation of blood and qi-invigorating chuanxiong, containing 1g drugs per milliliter. ③Normal saline group: 6 rats. estradiol was replaced by the same volume of normal saline, other useage was the same as that in the estradiol group.④Sham-operation group: 7 rats. without any drugs given. Rats were given the same treatments as that in the other group, but without fimbria-fornix transection. Somatosensory evoked potentials of cortex of all the animals were observed at 30 days after medication. RESULTS: All the 29 rats entered result analysis.①The amplitude of somatosensnry evoked potentials of cortex on the left side in the intervention group with Traditional Chinese medicine and normal saline group was significantly shorter than that in the sham-operation group [(27.34±8.50), (19.25±20.75).
