摘要
采用荧光光谱法和紫外-可见吸收光谱法研究了在模拟人体生理条件下不同温度时,血卟啉与人血清白蛋白结合反应的荧光猝灭光谱行为,根据Stern-Volmer方程和Lineweaver-Burk双倒数函数曲线计算了其结合常数在高温时要比低温时小;根据Fφster能量传递原理计算出结合位点r=4.36,并根据热力学参数确定了它们之间的作用力类型主要为氢键和VanderWaals力,HP进入了HSA的疏水腔。
The quenching reaction of hemoporphyrin with human serum albumin(HSA) was studied by using fluorescence spectra and absorption spectra. The formation constants of them were analyzed at different temperature according to Stem-Volmer equation and double-reciprocal equation, which are smaller at high temperature than at low temperature. The binding site was calculated ( r = 4.36) by Foster energy transfer mechanism, and the thermodynamic parameters were obtained. H-bond and Van der Waals are two main reactions between HP and HSA.
