摘要
目的:通过考察黄芩多糖对猪生殖和呼吸系统综合征病毒在传代细胞系MARC-145细胞中增殖的影响,证明黄芩多糖具有抑制病毒增殖作用。方法:感染病毒的细胞用不同浓度的多糖样品处理,处理后继续培养若干小时,进行台盼蓝染色、免疫荧光分析、蚀斑形成实验和聚合酶链式反应。结果:随着黄芪多糖浓度的提高蚀斑形成数减少,感染病毒细胞的死亡率降低,荧光分析阳性率下降,聚合酶链式反应产物条带减弱甚至消失。结论:黄芩多糖显示细胞水平抑制病毒增殖作用。
Objective:To show that polysaccharide from Scutellaria baicalensis Georgi has antiviral property by investigating its effect on replication of porcine reproductive and respiratory syndrome virus (PRRSV) in cells of the permanent MARC-145 cell line. Methods:Cells infected with virus were treated with various concentration of polysaccharide and incubated for some hours, then Trypan blue exclusive assay, immunofluorescence assay (IFA), plaque forming assay, polymerase chain reaction (PCR) was performed. Results: Number of plaques, the death rate of cells and percentage of IFA positive cells reduced and band of PCR product became weaker and undetectable with the concentration of polysaccharide increasing after infection with PRRSV. Conclusion:It was indicated that polysaccharide from Scutellaria baicalinsis Georgi could inhibit the replication of virus at cell level.
