摘要
综述PTEN作为一个重要的肿瘤抑制基因,其定位、基因结构、生物学功能、作用机制、表达调控机制、失活等方面的最新研究进展。PTEN通过PIP3途径、FAK途径、MAPK途径等发挥其脂质磷酸酶和蛋白磷酸酶活性,诱导细胞凋亡、抑制细胞生长、调节细胞迁移与黏附;参与胚胎的发育及维护免疫系统的稳定。PTEN的失活与肿瘤的发生紧密联系。PTEN蛋白的表达受p53、Egr-l、PPARγl等多种因子调控。
We summarize the latest investigation progress of PTEN(an important tumor suppressor gene)in its apposition, gene structure, biochemical functions, action mechanism, expression regulatory mechanisms, inactivations and so on. PTEN brings into its lipid and protein phosphatase activity through PIP3, FAK, MAPK pathway. PTEN also induces apoptosis, inhibites cell growth, regulates cell migration and adhesiveness. In addition, PTEN plays a pivotal role in embryonic development and the maintaince of immunie system. Furthermore, the inactivation of PTEN is associated with occurrence of tumor. The expression of PTEN protein is regulated by p53, Egr-1, PPAR71 and other factors.
