摘要
目的研究资木瓜皂苷(SCS)提取工艺的优化及对大鼠佐剂性关节炎(AA)的抗炎作用及免疫机制。方法以SCS的提取总量为考察指标,采取正交试验法考察乙醇浓度、溶媒用量、提取时间、提取次数等因素,每个因素设置3个水平。流式细胞术检测SCS对AA外周血中CD4+、CD8+的表达水平;MTT法检测SCS对AA外周T淋巴细胞增殖的影响。结果 SCS最佳提取工艺为乙醇浓度70%,溶媒容量10倍,提取时间1h,提取3次,提取率为1.7%。SCS能降低AA外周血中CD4+的表达水平,对CD8+的表达无明显影响,对AA外周T淋巴细胞增殖有抑制作用。结论最佳提取条件为A2B1C2D3,SCS通过抑制T淋巴细胞增殖和降低CD4+表达而发挥抗炎免疫作用。
OBJECTIVE To study optimization on extraction technology of saponin of Chaenomeles Speciosa( SCS) and study on its anti-inflammatory immunologic mechanism to rats adjuvant arthritis( AA) .
