摘要 采用聚合酶链反应(PCR)-单链构象多态性(SSCP)技术,分析了8例已知DR4等位基因型的个体,其中7例不同基因型的样本SSCP图谱有明显区别,2例均为DRB1*0410的样本,SSCP图谱则完全一致。笔者着重探讨了影响SSCP分析的主要因素:聚丙烯酰胺凝胶浓度和交联度,凝胶中是否含甘油,电泳温度、电流或电压大小等。提示PCR-SSCP可用于研究HLA的微观多态性、器官移植的配型及法医学上个体识别等方面,具有简便,快速、经济以及高度的分辨力和良好的重复性。 The technique of polymerase chain reaction(PCR)-single strand conformation polymorphism (SSCP) was adopted to analyse 8 samples of individuals whose HLA-DR4 alleles had been known. Among these ,7 samples with different genetic types had obvious distinction in SSCP band pattern. 2 of them were both samples of DRB1 0410 with the same SSCP pattern. The research emphysis was put on the main factors which influenced SSCP analysis concentration of polyacrylamide gel and degree of cross link; glycerol within the gel or not; electrophoresis temperature;current and voltage etc. It suggested that PCR-SSCP be available in studying the microscopic polymorphism,HLA matching of transplantation compatibility and individual recognition in forensic medicine etc. In addition,this technique is simple ,rapid,economical highly distinguishable and reproducible.
