摘要
本研究采用生物定量测定方法观察了豚鼠实验性哮喘时血浆lyso-pAF和肺泡灌洗液(BALF)中PAF的变化。结果显示:致敏组豚鼠哮喘发作时血浆lyso-PAF水平显著高于对照组(P<0.01),BALF中PAF水平也明显高于对照组(P<0.05),同时伴有血浆及BALF中TXB_2、TXB_2/6-酮-pGF1α水平的升高(P<0.01)。致敏+川芎嗪组豚鼠血浆lyso-pAF明显低于致敏组豚鼠(P<0.05),其BALF中PAF水平也有明显降低(P<0.05)。提示过敏性哮喘豚鼠发作期PAF产生增多,川穹嗪对PAF的产生具有一定的抑制作用。
Platelet-activating factor(PAF)is a potent inflammatory mediator.It wasdemonstrated that PAF may cause bronchoconstriction and increase vascular permeabilityin animal and human being.In order to investigate the role of PAF and its possiblemechanism in allergic asthma,the concentration of PAF in plasma and bronchoalveolarlavage fluid(BALF)of allergy asthmatic guinea pigs were observed.It was found that theplasma levels of lyso-PAF、TXB_2、TXB_2/6-keto-PGF_(1α) and the concentration of PAF,TXB_2、TXB ̄2/6-keto-PG_(1α),a in BALF increased significantly(P<0.01)in sensitized group,but 6-keto-PGF_(1α) decreased remarkably(P<0.05). However,in sensitized + ligustrazingroup,ligustrazin relieved the symptoms and inhibited the production of PAF,lyso-PAFand TXB_2 in plasma and BALF.It was concluded that PAF play an important role inallergic asthma and ligustrazin had some good effects in preventing experimental asthma.
