摘要
肿瘤细胞对化疗药物产生抗药性的原因有几种,其中最主要的是多药抗性基因(MDRI)过量表达。定量检测分析MDRI表达水平可说明病人的抗药能力的高低,判断预后,指导临床化疗。本研究通过逆转录PCR方法定量检测了20例大肠癌病人癌组织中MDRI的表达情况.其中包括10例未化疗过的病人.10倒多次化疗的病人.结果显示未化疗的病人MDR1的扩增量(平均为0.056)明显低于化疗后的病人(0.36),两者差异显著,表明MDR1扩增量与化疗密切相关.对临床化疗及判断预后有重要的参考价值。
One mechanism of drug resistance is associated with over expression of resistance gene(MDR1 )of the multidrug and phenotypic expression of transmembrance P-glycoprotein, which acts as an efflux pump resulting in decreased intracellular drug accumulation. In order to improve the therapeutic effect for individual patient,we devised a highly sensitive method to detect levels of MDR1mRNA in colorectal cancer cells based on PCR. Our results demonstrate that the expression of MDR1 may be correlated with the response of chemotherapy.
