摘要
Ⅳ型胶原片段７Ｓ是反映慢性肝病肝纤维化程度的重要血清学指标之一。本研究报道自人胎盘组织中提取、纯化该７Ｓ片段，并制备了高效价的兔抗血清。７Ｓ经ＳＤＳ—ＰＡＧＥ电泳鉴定为一均匀条带，分子量为３６０ｋＤ，氨基酸组成分析提示其成分与文献报道基本一致。经间接血凝试验、放射免疫法分析，自制的兔抗血清效价较高，特异性强，可用于放射免疫测定。
The 7S collagen was isolated from human placenta using pepsin solubisation followed by fractional salt precitation and chromatographic purification. Then the native preparation was characterized using amino acid analyses, disc gel electrophoresis, and immunological methods. The molecular weight detemined was about 360kD.7S collagen was immunogen and could be characterized by radioimmunoassays (RIA), indirect hemoagglutinatiion inhibition test. the rabbit antiserum against 7S collagen are comfortable for RIA.
