摘要
探索利用碱浸提和酶法去蛋白从赤魟软骨中提取分离纯化黏多糖的技术,并对该多糖进行了含量测定。结果表明:当NaOH浓度为6%,40℃提取6h,利用中性蛋白酶在40℃下酶解7h,三氯醋酸沉淀去蛋白,加入4倍体积的95%乙醇沉淀多糖时,黏多糖提取率可达21%以上,产品纯度约84.53%,白色粉末,同时对黏多糖的理化性质进行了鉴别。
The technique of extracting and refining mucopolysaccharide from Dasyatis akajei cartilage obtained was studied and the content of mucopolysaccharide was measured. The resuits showed that when the content of NaOH was 6%, temperature 40℃, extracting time 6h, followed neutral protein enzyme dissolution for 7h under 40℃, protein precipitation with trichloroacetic acid and 4 times volume 95% C2H5OH for polysaccharide precipitation, the extraction rate was over 21%,the product was white powder and its purity was about 84.53%. Physical and chemical properties of the mucopolysaccharide were identified.
