胰岛素对大鼠移植的胚胎脊髓组织存活与生长的影响 被引量:2

EFFECTS OF INSULIN ON SURVIVAL AND GROWTH OF TRANSPLANTED EMBRYONIC SPINAL CORD TISSUE IN RAT

摘要 用HRP逆行追踪技术和体视学方法,探讨了胰岛素对移植在缺损运动神经元的脊髓内的胚胎神经元存活与生长有无促进作用。将13~14d胚龄大鼠脊髓腹侧组织块,移植到受体大鼠左侧腰段缺损运动神经元的脊髓背外侧部。同时将受体鼠右侧带神经的 长伸肌移在左侧腰段脊椎旁,使其神经断端插入脊髓移植物的同一位置内。胰岛素组的胚胎移植物及覆盖其表面的硝酸纤维素膜,均经胰岛素生理盐溶液浸泡过,对照组仅用单纯生理盐溶液浸泡。术后大鼠存活6周。在移植的肌肉内注入HRP作逆行追踪。结果:1.胰岛素组胚胎移植物的体积为0.4252±0.0736mm3;对照组的为0.1383±0.0296mm3。2.胰岛素组移植物神经元胞体体积为0.0172±0.0041mm3;对照组的为0.0030±0.0011mm3。3.胰岛素组移植物神经元数目为5518±1343个;对照组的为1214±437个。4.胰岛素组移植物内和移植物邻近的受体脊髓组织中,可见HRP阳性神经元,而对照组则未观察到。以上结果提示:胰岛素能促进移植的胚胎脊髓神经元的存活与生长;有些神经元还支配了移植的肌肉,其中一些神经元很可能迁移到了受体脊髓组织中。 whether insulin could promote the survival and growth of embryonic spinal cord tissue transplanted into the rat spinal cord depleted motoneurons was explored by HRP retrograde tracing technique and stereological method.A small piece of rat spinal cord ventral tissue in embryonic days 13~14 was transplanted into the dorsolateral portion of host's rat left lumbar spinal cord depleted motoneurons.In the same time,the right extensor hallucis longus(EHL)muscle of the host's rat with its nerve was placed between left lumbar vertebral column and paravertebral muscle. The nerve's proximal end was inserted into the same place which had accommodated the transplant.In insulin group,the embryonic graft and the nitrocellulose covering the surface of graft were immerged in insulin saline solution before transplantation,While in control group those were immerged in pure saline solution,The postoperative rats survived for 6 weeks,HRP was injected into the implanted EHL muscle for retrograde tracing.The results showed as follows;1.The graft's volumn was 0.4252±0.0736 mm3 in insulin group;that was 0.1383±0.0296 mm3 in contro group.2.The volume of neuronal soma at the graft was0.0172±0.0041 mm3 in insulin group;that was 0.0030±0.0011 mm3 in control group.3.The neuron's number at the graft was 5518±1343 in insulin group;that was 1214 ± 437 in control group.4.HRP positive neurons were observed at the graft and at host's spinal cord tissue adjoining the graft in insulin group,but those were not seen in control group.These results suggested that insulin could promote the survival and growth of neurons in embryonic spinal graft,the reinnervation of the some neurons to the implanted target muscle and the immigration of a few of among neurons into host's spinal tissue.
