碱性成纤维细胞生长因子对大鼠脑出血血肿周围脑组织和血浆中血栓素B_2和6-酮-前列腺素F_(1a)含量的影响

摘要 目的观察大鼠脑出血后应用碱性纤维细胞因子(bFGF)对血肿周围脑组织及血浆中TXB2和6-K-PGF1a含量及比值的影响。方法采用脑内注射胶原酶建立大鼠脑出血模型,分为脑出血模型对照组和bFGF治疗组,并于脑内注射生理盐水建立假手术组对照组。每组又分为1d和3d两个时间点,bFGF治疗组应用bFGF进行治疗,检测1d和3d后血肿周围脑组织及血浆中TXB2和6-K-PGF1a含量。结果bFGF治疗组与模型对照组比较,1d时血浆中TXB2含量降低(P<0.05),1d和3d时6-K-PGF1a含量则均升高(P<0.05),TXB2/6-K-PGF1a比值1d(P<0.05)和3d(P<0.01)时均降低;bFGF治疗组与模型对照组比较,血肿周围脑组织中TXB2含量3d时较脑出血模型组升高(P<0.01),6-K-PGF1a含量无显著差异(P>0.05),TXB2/6-K-PGF1a比值1d时降低(P<0.05),3d时升高(P<0.05)。结论脑出血后血浆中TXB2和6-K-PGF1a含量的变化比血肿周围脑组织中明显;bFGF可使大鼠脑出血后血浆中TXB2含量减少,6-K-PGF1a含量增多,TXB2/6-K-PGF1a比值降低。 Objective To study the change and the ratio of concentration of TXB2, 6-K-PGF1, in perihematoma brain tissue and plasma in rat with intracerebral hemorrhage,and the effect of basic fibroblast growth factor(bFGF). Methods The rat model of experimental intracerebral hemorrhage(ICH) was established by injection of collaginaseW into internal capsule of cerebrum. The animals neuroethological scores more than 3 by Bederson method after the operation were enrolled in the study, and SO control group was established by injection of physiological saline. 48 Sprague-Dawley rats were randomly divided into SO control group , model of ICE group and bFGF treatment group. Each group was subdivided into first day and third day group. The concentration of TXB2 and 6-K-PGF1a in perihematoma brain tissue and plasma were measured. Results Compared with model of ICE group, the concentration of TXB2 in plasma of bFGF treatment group decreased at first day(P〈0.05),and the concentration of 6-K-PGF1, increased both at first day and third day(P〈0. 05). The ratios of TXB2/6-K-PGF1a, decreased both at first day(P〈0. 05) and third day(P〈0. 01), while, the concentration of TXB2 in perihematoma brain tissue of hFGF treatment group increased at third day(P〈0. 01) ,there was no significant change of the concentration of 6-K-PGF1a, (P〉0. 05), and the ratios of TXB2/6-K-PGF1a. decreased at first day(P〈 0. 05) and increased at third day(P〈0.05). Conclusion The change of concentration of TXB2 ,6-K-PGF1, in plasma is more significant than that in perihematoma brain tissue. The use of hFGF can decrease the concentration of TXB2 in plasma of rat with intracerehral hemorrhage , hut can increase the concentration of 6-K-PGF1a, then the ratio of TXB2/6-K-PGF1, decreases.
