非小细胞肺癌Bcl-2及Bax蛋白的表达及其与细胞凋亡和增殖的关系 被引量:11

摘要 ①目的探讨非小细胞肺癌Bcl-2、Bax蛋白的表达及其与细胞凋亡和增殖的关系。②方法采用SABC免疫组化法检测43例非小细胞肺癌及20例癌旁正常肺组织Bcl-2、Bax蛋白及增殖细胞核抗原(PCNA)表达水平;用TUNEL法检测细胞凋亡情况。③结果肺癌组织Bcl-2蛋白的表达显著高于癌旁肺组织(χ2=4.74,P<0.05);肺癌组织Bax蛋白的表达显著低于癌旁肺组织(χ2=5.65,P<0.05);肺癌组织中细胞凋亡指数(AI)和增殖指数(MI)均明显高于癌旁正常肺组织(t=8.28、9.62,P<0.01);Bcl-2蛋白表达阳性的肺癌组织AI明显低于表达阴性组(t=2.41,P<0.05);Bax蛋白表达阳性的肺癌组织AI明显高于表达阴性组(t=2.62,P<0.05);Bcl-2、Bax蛋白的表达均与MI无关(t=0.70、0.22,P>0.05)。④结论Bcl-2蛋白具有抑制肺癌细胞凋亡作用,而Bax蛋白具有促进肺癌细胞凋亡作用,因肺癌组织存在Bcl-2蛋白表达上调,Bax蛋白表达下调,导致细胞增殖和凋亡失衡,在肺癌发生、发展过程中可能具有重要作用。 Objective To study the expression of Bcl-2 and Bax oncoprotein and its relationship with proliferation and apoptosls in non-small-cell lung cancer (NSCLC). Methods Immunohistochemistry stain (SABC) method was applied to investigate the expression of Bcl-2, Bax protein and PCNA in 43 NSCLC and 20 normal lung tissue adjacent to lung carcinoma. Cell apoptosis was examined with TUNEL method. Results The positive rate of Bcl-2 was significantly higher (x^2=4.74, P〈0.05), while that of Bax was significantly lower (x^2=5.65,P〈0.05) in the tissue of patients with NSCLC than in normal lung tissue adjacent to carcinoma. The AI and MI were higher than those of normal lung tissue adjacent to lung carcinoma (t=8.28,9.62;P〈0.01). The AI of cancer patients with positive Bcl-2 was obviously lower than that with negative Bcl-2 (t=2.41,P〈0.05). AI of Bax-positive cancer patients was obviously higher than that of Bax-negative ones (t=2.62, P〈0.05). The expression of Bcl-2 and Bax oncoprotein had no relationship with MI of lung cancer (t=0.70,0.22;P〉0.05). Conclusion The expression of Bcl-2 protein is high while that of Bax protein is low in NSCLC. The expression of Bcl-2 and Bax protein correlates with apoptosis, which might play an important role in the carcinogenesis of lung carcinoma.
