摘要 目的:研究过量表达ErbB2对于MCF-7细胞生长和侵袭的作用。方法:构建带有ErbB2逆转录病毒,感染MCF-7细胞,检测ErbB2的表达。用转染后的细胞开展体外生长和侵袭实验。结果:转染后ErbB2在MCF-7细胞中过量表达。体外生长和侵袭实验显示,过量表达ErbB2的细胞增殖快、侵袭性强。结论:过量表达ErbB2促进MCF-7细胞的生长和侵袭;阻断ErbB2信号可能为高表达ErbB2的肿瘤治疗带来新的策略。 AIM: To investigate the effects of ErbB2 overexpression on growth and invasiveness in cultured MCF - 7 cell line. METHODS: Retrovirus containing ErbB2 gene was transfected into MCF - 7 cells and ErbB2 expression was detected by Western blotting. Proliferation and invasive assays were carried out. Cells overexpressed ErbB2 and its control, AP2, were used in the experiment. RESULTS: ErbB2 was overexpressed in MCF - 7 cells after tiansfection. In vitro, cells overexpressed ErbB2 showed highly proliferated and highly invasive characteristics compared to the control cells. CONCLUSION: Overexpression of ErbB2 promotes cell proliferation and enhances invasiveness in MCF - 7 cells. Inhibition of signaling induced by ErbB2 might be a novel strategy for the therapeutics of cancer with ErbB2 overexpression.
