摘要
目的:观察对生物膜具有保护作用的携硒、锌、维生素E大豆磷脂脂质体对实验性大鼠心肌缺血损伤的影响。方法:实验于2002-06/2003-09在武汉大学医学院病理生理实验室完成。实验大鼠40只,单纯心肌缺血组10只、大豆磷脂脂质体保护组10只、携硒和锌及维生素E的大豆磷脂脂质体保护组10只,皮下注射异丙基肾上腺素30mg/kg,连续3d,制成心肌缺血模型;正常对照组10只不造模,注射等量的生理盐水。造模前大豆磷脂脂质体保护组灌服大豆磷脂脂质体(150mg/kg),携硒和锌及维生素E的大豆磷脂脂质体保护组大鼠灌服携硒、锌、维生素E的大豆磷脂脂质体20d,灌胃量为10μL/g,正常对照组、单纯心肌缺血组则灌服等量的生理盐水。检测大鼠血浆磷酸肌酸激酶活性采用生化法,检测心肌细胞凋亡率应用脱氧核糖核苷末端转移酶标记法,检测细胞凋亡相关基因Bcl-2及Bax表达应用免疫组织化学法。结果:进入结果分析实验大鼠40只。①血浆磷酸肌酸激酶活性:单纯心肌缺血组比正常对照组显著增强犤(4.4±0.8),(2.2±0.4)μkat/L,P<0.01犦。大豆磷脂脂质体保护组较单纯心肌缺血组明显降低犤(3.2±1.3)μkat/L,P<0.01犦,而携硒和锌及维生素E的大豆磷脂脂质体保护组也较单纯心肌缺血组显著降低犤(2.4±0.5)μkat/L,P<0.01犦,但两处理组之间无差异。②心肌细胞凋亡率:单纯心肌缺血组明显高于正常对照组犤(31.81±5.63)%,(3.59±1.35)%,P<0.01犦,大豆磷脂脂质体保护组及携硒和锌及维生素E的大豆磷脂脂质体保护组明显低于单纯心肌缺血组犤(19.91±4.03)%,(13.33±2.79)%,P<0.01犦,且后者减少缺血心肌细胞凋亡的效应明显高于单纯大豆磷脂脂质体处理组(P<0.05)。③心肌组织Bcl-2及Bax表达(吸光度值)的比较:单纯心肌缺血组均比正常对照组表达明显增多(0.302±0.017和0.281±0.011,0.304±0.013和0.253±0.011,P<0.01或0.05);大豆磷脂脂质体保
AIM: To observe the influence of soybean phospholipid liposome, which enwraps selenium, zinc and vitamin E and has a protective effect on myocardial ischemia injury of rats. METHODS: The experiment was carried out in the pathophysiological laboratory, Medical College of Wuhan University between June 2002 and September 2003. Forty experimental rats were used: Rats in the simple myocardial ischemia group (n=10), soybean phospholipid liposome protection group (n=10) and soybean phospholipid liposome enwrapped selenium, zinc and vitamin E protection group (n=10) were made into models of myocardial ischemia by subcutaneous injection of isoprenaline (30 mg/kg) for 3 continuous days, and those in the normal control group were not made into models and injected with saline of the same volume. Before model establishment, rats in the soybean phospholipid liposome protection group and soybean phospholipid liposome enwrapped selenium, zinc and vitamin E protection group were perfused with soybean phospholipid liposome (150 mg/kg) and soybean phospholipid liposome (10 μL/g for 20 days), which enwrapped selenium, zinc and vitamin E protection, and those in the normal control group and simple myocardial ischemia group were perfused with saline of the same volume. The activity of creatine phosphokinase in plasma was detected with biochemistry, the myocardial apoptotic rate was detected with terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL), the expressions of apoptosis related genes were determined with immunohistochemistry. RFSULTS: All the 40 experimental rats were involved in the analysis of results. ① The activity of creatine phosphokinase in plasma: It was significantly enhanced in the simple myocardial ischemia group as compared with the normal control group [(4.4±0.8), (2.2±0.4) μkat/L, P〈 0.011; It was obviously decreased in the soybean phospholipid liposome protection group ascompared with the simple myocardial ischemia group [(3.2±1.3) μk
