摘要
目的研究银杏叶提取物EGb761对肿瘤坏死因子-α(TNF-α)诱导HeLa细胞凋亡和Caspase-8活性的影响.方法采用流式细胞术检测细胞凋亡,Caspase-8荧光检测试剂盒测定Caspase-8活性.结果EGb761在10~40 mg/L终浓度范围内对rhTNF-α诱导的HeLa细胞凋亡均有显著的抑制作用(P<0.01),呈剂量依赖关系;EGb761在10~40 mg/L终浓度范围内对rhTNF-α诱导的HeLa细胞Caspase-8活化均有显著的抑制作用(P<0.01),呈剂量依赖关系.结论结果提示,EGb761抑制TNF-α诱导的HeLa细胞凋亡可能与其抑制Caspase-8活化有关.
Objective To investigate the effects of Ginkgo biloba extract EGb761 on apoptosis of HeLa cell induced by tumor necrosis factor-α. (TNF-α.) and on the activity of Caspase-8. Methods The effect of EGb761 on the apoptosis of HeLa cell was induced by rhTNF-α and Caspase-8 fluorescent kits was used to detect the activity of Caspase-8. Results The result of flow cytometry exhibited that EGb761 with 10 ~ 40 mg/L final concentration had notable inhibition but not absolute inhibition on the apoptosis of HeLa cell induced by rhTNF-α ( P 〈 0.01 )and it showed dose-dependence. The detection of the activity of Caspase-8 showed that EGb761 with 10 ~ 40 mg/L final concentration also had notable inhibition on the Caspase-8 activation of HeLa cell induced by rhTNF-α ( P 〈 0.01 ). Conclusion The inhibition of apoptosis in HeLa cell inducedby rhTNF-α by EGb761 may have relation to the Caspase-8 activation.
