摘要
目的研究树突状细胞疫苗瘤内注射对胶质瘤的治疗作用。方法建立大鼠C6脑胶质瘤皮下和颅内肿瘤模型,通过第一组为瘤内注射树突状细胞(DC)、第二组为腹部皮下注射DC、第三组为对照组,观察肿瘤生长情况和大鼠生存时间,比较细胞毒性T淋巴细胞(CTL)的活性,并进行病理学检查。结果治疗组大鼠表现为肿瘤生长部分抑制,生存时间明显延长,与对照组比较,有极显著差异(P<0.01),瘤内注射DC和腹部皮下注射C6冻融抗原致敏DC,两治疗组间无显著性差异(P=0.60);CTL杀伤活性增强,和对照组比较有显著性差异(P<0.05)。双侧皮下种植的肿瘤治疗一侧后,生长均受到抑制,并逐渐缩小。结论瘤内注射DC治疗脑胶质瘤是一种新的、有效的免疫治疗策略。
Objective To evaluate intratumoral injection of dendritic cells (DC) for the treatment of malignant gliomas. Methods With using subcutaneous and intracranial C6 brains glioma models of rats, the growth of the tumors and survival time of the rats were observed 'after the application of intratumoral injection of dendritic cells.The brains were removed and examined pathologically when the rats died. Results The intmtumoral injection of dendritic cells resulted in partially tumor regression in the treated animals. The survival time increased which were statistically significant (P 〈0.01 ), but there was no statisticall difference between the two experimental groups (P =0. 60). The antitumor cytotoxic T-lymphocyte(CTL) activity increased (P 〈0.5). Bilateral tumors were inoculated in both lower flanks. Unilateral tumor was treated hy DC injection and both tumors were depressed. Conclusion These novel findings demonstrate intraturnoral injection of DC as an effective therapeutic strategy for intracranial malignant gliomas.
