摘要
目的 通过立体定向技术建立大鼠脑原位移植瘤模型,观察其生长特性。方法 在立体定向仪引导下,将体外培养的大鼠C6胶质瘤单细胞悬液(1×10 6/10 μl)接种于2 7只Wistar大鼠右尾状核区。以接种C6细胞后大鼠的允许生存期,将大鼠分成2组,观察大鼠生存状态、成瘤情况、肿瘤体积、脏器转移灶、组织学形态、胶质纤维酸性蛋白(GFAP)、血管内皮生长因子(VEGF)、兔抗人因子Ⅷ相关抗原免疫组化染色。结果 大鼠成瘤率为10 0 % ,无颅外转移。荷瘤大鼠平均生存期为(2 5 5 0±5 75 )d。肿瘤体积在第3~4周时肿瘤迅速增大。肿瘤呈浸润性生长,甚至侵袭颅骨;瘤细胞病理性核分裂像多见,GFAP蛋白呈散在阳性表达,VEGF蛋白强阳性表达;瘤组织内有典型的栅栏状坏死和丰富的微血管。结论 C6细胞在大鼠脑内原位移植瘤与人脑胶质母细胞形态和生长特性十分相似,具有分化程度低、微血管丰富、生长速度快且高度侵袭等特点。
Objective To establish an orthotopic implantation model of C6 cells in Wistar rats with stereotactic technique and to study the growth patterns of the tumors formed in the rat brain. Methods The C6 cells (10 6/10 μl) were stereotaticallly implanted into the right caudate nuclei of 27 Wistar rats. All rats were divided into two groups according to their survival time. After C6 inoculation, the living and survival states of rats with glioma were observed and the formation and their volumes of tumors, metastasis, histopathology, expression of glial fibrillary acidic protein (GFAP), vascular endothelial growth factor (VEGF) and factor Ⅷ related antigen in the tumors were investigated. Results All the tested rats developed brain tumors without extracranial metastasis. The mean survival of the tumor-bearing rats was (25.50±5.75) d. The tumor volumes increased in a time-dependent manner and reached maximum between 3 to 4 weeks. The tumors were highly cellular, with remarkable cytoplasmic and nuclear polymorphism, intratumoral pseudopalisade necrosis, mitotic figures and abundant vessels which are immunostained with anti-factor Ⅷ related antigen polyclonal antibody. Immunohistochemical staining showed the distribution of VEGF protein was mainly in tumor cells of malignant glioma. The tumor margins displayed an invasive zone with tumor cells infiltrating the surrounding normal brain tissue matrix. The GFAP positive expression of glioma showed glial-derived tumor. Conclusion The orthotopic tumors formed by C6 cells in the rat brain shared similarities with human glioblastoma in morphology and differentiation, which could be used for experimental therapeutic studies on malignant gliomas.
