摘要
采用反相高效液相色谱法同时测定了洗必泰及有关杂质对氯苯胺浓度。用Zorbax-C_8柱,0.2mol/LNaH_2PO_4(pH3.0)-甲醇(49.5:50.5)为流动相,非那西丁为内标,240nm波长检测。本法简便准确,重现性好。洗必泰和对氯苯胺分别在15~200μg/ml和60~1200μg/ml浓度范围内线性关系良好(r=0.9999),最低检测限分别为1.33μg/ml和7.5μg/ml(S/N=3:1),平均回收率分别为100.1%和103.1%。相对标准偏差R50%分别为1.41%和1.14%。
This
paper reports a new RP-HPLCmethod using phenacetin internal standard(IS)for si-multaneous
determinatiOn of Chlorhexdine(CH)andits impurity 4-chloroaniline ( 4-CA). We use the LC-3A
HPLC instrument,a Zorbax-C_8 column(25cm ×4. 6mm id , 10μm),a variable wave length UV
detectorand C-RIA chromatographic data processor。A mix-ture of methanol and 0. 2mol/L
NaH_2PO_4(50.5:49. 5,pH3. 0) is used as mobile phase, whose flow rateis 1. 0ml/min. Sample
solution of 15μl is injected ontothe column,which is maintained at 30℃.The eluent ismonitored
at 240nm.The retention times are 16.72minfor CH and 5.54min for 4-CA.The standard curvesof
CH and 4-CA are established using peak area ratioof CH/IS and 4-CA/IS to concentration
respectively.They both are linear (r=0.9999)within 15~200μgml for CH and 60~1200μg/ml
for 4-CH. The limits ofdetection are l. 33μg/ml for CH and 7. 5μg/ml for 4-CA ( S/N=3). The
analytical recovery rates are100.1%for CH and 103.1%for 4-CA.This methodhas been used for
investigating the stability of CHpreparatioon.
