摘要
应用全细胞斑片钳技术记录酶学方法分离的豚鼠单个心室肌细胞离子通道电流。通过改变计算机内PClamp软件包(7.71版本,美国Axon公司)中有关参数,设置钠通道电流(Ina)及INa动力学研究的脉冲刺激方案。研究显示,INa表现电压和时间依赖性,本实验条件下所引导的INa能稳定地用于对钠通道灭活特性,使用依赖性阻滞以及通道灭活后恢复过程等动力学研究。本文对钠通道有关动力学特征及其意义作了初步探讨。
Transmembrane ionic channel currents were recorded by using the whole-cell patchclamp technique in single cardiac ventricular myocyte enzymatically isolated from guinea-pig hearts.The pulse-stimulating protocols for sodium channel current(INa)and its kinetic studies are designed by changes in related parameters in PCl amp software(7.71 version,USA.Axon INc).The results showed that INa is characteristic of voltage-and time-dependence;INa recorded in this experimental condition can be well used for kinetic studies of sodium channel inactivation, use-dependent blocking,and recovery course from blocking.
