摘要
实验采用ABC免疫组织化学方法研究了八肽胆囊收缩素样免疫反应(CCK-8-IR)物质在猪、大鼠和豚鼠肠道的定位与分布,并用相邻切片免疫双标记法观察了它与5-羟色胺(5-HT)的关系。结果表明,CCK-8-IR细胞主要位于肠腺的底部,少数位于绒毛上皮。节段性分布上,在猪可见于从十二指肠到结肠全长的粘膜,大鼠和豚鼠CCK-8-IR细胞则见于从十二指肠至回肠的粘膜,但均以十二指肠密度最高;免疫双标记法证实,在三种动物肠道中均有CCK-8与5-HT共存的现象。
Cholecystokinin (CCK), a peptide with multiple molecular forms and physiological functions, is widely distributed in both the nervous system and the endocrine cells of digestive tract. In this study, the localization and distribution of cholecystokinin octapeptide (CCK8) in the porcine, rat and guinea pig intestinal tract were investigated by ABC immunohistochemical technique, and the possible coexistence of CCK-8 with serotonin was also observed by using adjacent section double immunolabelling method. It was found that the CCK-8 immunoreactive (CCK-8-IR) cells were maidy localized in the lower parts of intestinal glands, while only a few could be observed on the epithelium of villi. In the aspect of segmental distribution. the CCK-8-IR cells could be seen in the mucosa of the whole intestinal tract from duodenum to colon in pig, while in rat and guinea pig, the CCK-8-IR cells were only seen in the mucosa from duodenum to ileum. But, in the aspect of quantity, the density of CCK-8-IR cells showed the highest number only in duodenum. Nearby the adjacent sections of the duodenum, the results obtained by double immunolabelling method proved the coexistence of CCK-8 and serotonin in the intestinal tracts of all three animals.
