米非司酮对子宫腺肌病NF-κB和细胞因子表达的影响 被引量:23

摘要 目的:研究米非司酮和三苯氧胺对子宫腺肌病核因子κBp65mRNA(NFκBp65mRNA)、NFκB、血小板衍生生长因子A(PDGFA)和转化生长因子beta(TGFbeta)表达的影响。方法:子宫腺肌病妇女46例,随机分为米非司酮组、三苯氧胺组和对照组,药物治疗3个月。应用原位杂交法检测NFκBp65mRNA的表达;应用免疫组化法检测PDGFA,TGFbeta,NFκB的表达。结果:NFκBp65mRNA、NFκB、PDGFA和TGFbeta的表达强度,依次为对照组高于三苯氧胺组,三苯氧胺组高于米非司酮组,3组存在显著性差异。NFκB与PDGFA、TGFbeta表达呈正相关。结论:米非司酮和三苯氧胺抑制子宫腺肌病NFκBp65mRNA和NFκB的生成,降调PDGFA、TGFbeta表达。 Objective: To investigate the effects of Mifepristone and Tamoxifene on the expression of NF-κ B p65 mRNA, NF-κ B, PDGF-A and TGF-β of the adenomyosis. Methods: 46 cases of adenomyosis were randomly divided into 3 groups: Mifepristone group, Tamoxifene group, and the control group. Cytokines NF-κ B p65 mRNA was measured by ISH. The expression of NF-κ B, PDGF-A, and TGF-β was assayed by immunohistochemistry. Results: The expression intensity of NF-κ B p65 mRNA ,NF-κ B,PDGF-A and TGF-β was higher in control group, orderly decreased to Tamoxifene group, and then Mifepristone group, there was significant difference among them. The expression of NF-κ B was positive correlation with the expression of PDGF-A and TGF-β, respectively. Conclusions: Mifepristone and Tamoxifene could inhibit the production of NF-κ B p65 mRNA and NF-κ B in adenomyosis, and induced the down regulation of the expression of PDGF-A and TGF-β also.
