摘要
目的:探讨迷路炎时,豚鼠耳蜗IL - 2、TNF -αmRNA的表达变化及其与迷路炎的关系。方法:随机将2 4只豚鼠分为实验组和对照组,实验组18只、36耳,将1mg .ml- 1内毒素(lipopolysaccharide ,LPS) 0 .2ml缓慢注入中耳腔,对照组6只、12耳注入等量生理盐水,观察6h、4 8h、14d耳蜗的病理变化;用原位杂交技术检测IL - 2、TNF -αmRNA在耳蜗的表达,用Tiger92 0图像分析软件分析图象。结果:(1)耳蜗形态学改变:鼓室注药后6h ,实验组豚鼠耳蜗的血管纹、螺旋韧带、蜗螺旋轴静脉等即可见充血、水肿、炎细胞浸润,Corti器支持细胞和感觉细胞轻度肿胀、变性;4 8h时加重;14d时耳蜗各部分结构恢复正常。对照组豚鼠耳蜗各部分结构无改变;(2 )IL - 2mRNA的表达:对照组耳蜗无IL - 2mRNA表达;鼓室注射LPS后6h ,实验组耳蜗的螺旋神经节、螺旋缘呈阳性表达,血管纹、螺旋韧带呈可疑阳性;而4 8h、14d无表达;(3)TNF -αmRNA的表达:对照组耳蜗无TNF -αmRNA表达;鼓室注射LPS后6h ,TNF -αmRNA广泛表达于实验组耳蜗的螺旋神经节、Corti器、螺旋缘、血管纹、螺旋韧带等部位,4 8h表达明显增强(P <0 .0 1) ,14d时仅螺旋神经节呈弱阳性染色。结论:IL - 2、TNF -α可能与急性迷路炎的发生有关,TNF -α则可能起着更为重要的作用。
Objective:To investigate the relationship between the expressions of IL-2 ,TNF-α mRNA in cochlea and labyrinthitis in guinea pigs.Methods: Twenty-four pigmented guinea pigs were randomly allocated into two groups. 0.2 ml lipopolysaccharide(LPS) (1.0 mg. ml -1) or saline were transtympanically injected in experimental group (n=18) and control group (n=6) respectively. The morphology changes of cochleas were observed at 6 hours, 48 hours and 14 days after the application transtympanically. The expressions of IL-2 and TNF-α mRNA were detected through the technique of hybridization in situ and computer image analysis system.Results: (1) Pathomorphology change of inner ear: remarkable reaction such as inflammatory infiltration, cell degeneration, hyperemia, edema and disorganization were observed in the spiral ligaments(SL), stria vascularis (StV) and hair cell at 6 hours and 48 hours, but all the above mentioned signs disappeared at 14 days in experimental group. However, no abnormality was seen in the labyrinths in control group;(2) expression of IL-2 mRNA: in control group, expression of IL-2 mRNA in guinea pigs′ cochlea could not be viewed. In experimental group, the positive staining was observed in spiral ganglion cells (SGC) and spiral limbus and probable positive staining was observed in the SL and StV at 6 hours, while negative expression existed at 48 hours and 14 days; (3) expression of TNF-α mRNA: TNF-α mRNA was not expressed in the guinea pigs′ cochlea in control group, however, it was generally expressed in the SGC, Corti's organ and spiral limbus, while some weak positive cells were observed in the StV, SL and spiral modiolar vein (SMV) at 6 hours after LPS stimulation. The immunostaining was more significant at 48 hours than that at 6 hours (P<0.01) in experimental group, however, only SGC was weakly stained at 14 days. Conclusion: Acute labyrinthitis may be related with the activation of IL-2 and TNF-α mRNA, while the activation of TNF-α mRNA may be play a more important role.
