感染性神经性耳聋动物模型的建立及评估

Evaluation and establishment of animal model of infective sensorineural hearing loss

摘要 目的:神经性耳聋的发病机制不明,疗效不佳,建立G-菌内毒素所致感染性神经性耳聋动物模型,为该类疾病的研究奠定基础。方法:实验于2003-07/2004-07在创伤、烧伤与复合伤研究国家重点实验室完成。选择健康杂色豚鼠48只,根据脂多糖不同浓度随机分为0.01,0.1,1g/L脂多糖组及对照组。实验组动物鼓室内注入0.2mL,对照组注入等量生理盐水,注药后6,48h,2周,做听性脑干诱发电位(auditorybrainstemresponse,ABR)测试,并处死动物,观察中耳腔、耳蜗、内淋巴囊的病理变化。结果:①ABR结果:与对照组相比,0.01,0.1g/L脂多糖组豚鼠听阈在整个观察期内变化不明显(P>0.05);1g/L脂多糖组听阈显著增高(t=-3.28,-10.9,P<0.01),48h达高峰,2周时恢复正常。②中耳、内耳的形态学改变:0.01g/L脂多糖组中耳、内耳无炎性改变;0.1g/L脂多糖组部分动物6和48h中耳有炎性表现但内耳尚未见异常;1g/L脂多糖组6h已有明显中耳腔炎性反应,同时有内淋巴囊囊周、血管纹和螺旋韧带充血水肿、炎细胞浸润,Corti器毛细胞变性等内耳组织的病理改变;并在48h达高峰,2周中耳、内淋巴囊及耳蜗炎症逐渐消退。结论:鼓室给药是建立感染性神经性耳聋动物模型的有效途径,所建模型稳定、可靠,方法易于掌握;1g/L脂多糖是建立该模型的最佳浓度。 AIM:To evaluate and establish the experimental animal model of infective sensorineural hearing loss induced by bacterial endotoxin attributing to its unclear pathogenesis and poor effect,so as to provide a basis for the research of such disease. METHODS:The experiment was conducted in the National Key Laboratory of Wound,Burn and Combined Injury during July 2003 and July 2004.Forty eight pigmented healthy guinea pigs were randomly divided into four groups according to different concentration of lipopolysaccharide(LPS):LPS 0.01 g/L group,LPS 0.1 g/L group,LPS 1 g/L group and control group.In three experimental animals,three different concentrations(0.01,0.1,or 1.0 g/L) of LPS were transtympanically injected 2 mL respectively.The equal volume of saline was transtympanically injected in each animal of control group.The auditory brainstem response(ABR) thresholds were recorded at 6,48 hours and 14 days after the application of LPS.Then the detected animals were killed.The morphological changes of middle ear cavity,cochlea and endolymphytic sacs(ES) were observed. RESULTS:① The results of ABR thresholds:The ABR thresholds of the animals in the LPS 0.01 and 0.1 g/L groups were insignificantly changed,as compared with that in the control group(P >0.05),while significantly increased in the LPS 1 g/L group(t=- 3.28,- 10.9,P< 0.01,peak at 48 hours and normal level at 2 weeks after the application of LPS) during the whole observation period.② The morphological changes of middle ear and inner ear:Inflammatory reaction was not observed in the animals' middle ears and inner ears in LPS 0.01 g/L group.Although the middle ears had inflammatory response in some animlas,no abnormality was seen in the inner ears of aminals in LPS 0.1 g/L group at 6 and 48 hours.In 1 g/L group,remarkable inflammation existed in the middle ears,meanwhile,those signs such as inflammatory infiltration,Corti hair cell degeneration,hyperemia,edema and disorganization were observed in the spiral ligaments,stria vascularis,and perisaccular region o
