摘要
目的探讨低氧预适应(8%O2,3h)对大鼠大脑中动脉缺血2h再灌注后12h神经细胞凋亡及Caspase-3活性的影响。方法低氧预适应后12h制作脑缺血再灌注模型,大鼠大脑中动脉阻塞2h,灌注损伤12h,用TUNEL法和荧光四肽底物法分别检测假手术组、缺血再灌注组和低氧预适应组的凋亡细胞百分率和Caspase-3活性水平。结果与缺血再灌注组比较,低氧预适应组和假手术组的凋亡细胞百分率和Caspase-3活性水平显著降低(P<0.05,P<0.01);低氧预适应组与假手术组比较,细胞凋亡率明显升高(P<0.01),Caspase-3水平反而降低(P<0.01)。结论低氧预适应可减少大脑中动脉缺血2h再灌注后Caspase-3活性,抑制神经细胞的凋亡。
Objective To study the effect of hypoxic preconditioning in 8% O2 for 2 h on the rat brain nervous cell apoptosis and activity of Caspase-3 after rat cerebral ischemia-reperfusion injury.Methods Rats were subjected to hypoxic preconditioning for 12 h to develop cerebral ischemia-reperfusion model,the middle arteries of rats were occluded for 2 h,and reperfused for 12 h the apoptotic cell rate and Claspase-3 activity level were determined in the sham-operated group ,the ischemia-reperfusion group and the cerebral ischemia-reperfusion group by using TUNEL and AC-DEVD-AMC, respectively. Results The apoptotic cell rate and Caspase-3 activity level were significantly lower in the sham-operated group and the cerebral ischemia-reperfusion group than those in the ischemia-reperfusion group(P<0.05,P<0.01).Compared with the sham-operated group,the apoptotic cell rate rose dramatically and the Caspase-3 activity level were lower in the cerebral ischemia-reperfusion group(P<0.01).Conclusion One of the mechanisms of brain protection by hypoxic preconditioning may be due to the decrease of neural cell apoptosis and the activity level of Caspase-3.
