环孢素A对感染旋毛虫小鼠的免疫调节作用 被引量:1

摘要 目的观察环孢素A(CsA)对感染旋毛虫小鼠免疫功能的影响。方法小鼠腹腔注射环孢素A感染旋毛虫后,分别采用ELISA和流式细胞仪检测小鼠不同时期外周血中IL-2含量、CD4+及CD8+T淋巴细胞的百分率。结果(1)单纯感染旋毛虫小鼠感染后1~5周IL-2的含量较正常组明显增加;注射CsA组小鼠感染后1周IL-2的含量较单纯感染组明显降低,感染后2~4周IL-2的含量与单纯感染组相比无明显差异,但仍高于正常组。(2)单纯感染旋毛虫小鼠感染后1~4周,CD4+T淋巴细胞较正常组明显减少,CD8+T淋巴细胞明显增多,CD4+/CD8+比值明显下降;注射CsA组小鼠CD4+T淋巴细胞细胞较单纯感染组明显减少,而CD8+T淋巴细胞无明显变化。结论CsA对感染旋毛虫小鼠具有一定的免疫调节作用。 The effect of Cyclosporin A on the immune functions of mice infected with Trichinella spiralis was investigated in the present study, in which mice were infected with T.spralis, and Cyclosporin A (CsA) was given intraperitoneally 11 days after infection. At different periods, the content of IL-2 in peripheral blood was determined by ELISA,and the percentages of CD^+_4 and CD^+_8 cells in peripheral blood were tested by using flow cytometry. It was demonstrated that the content of IL-2 in peripheral blood of mice 1-5 weeks after infection was significantly increased than in the normal mice, but it decreased significantly in CsA treated group of mice one week after infection, and there was no difference between the infection group and the CsA-treated group of mice during 2-4 weeks.Meanwhile, the percentage of CD^+_4 cells in the infection group was significantly decreased in comparison with that of normal mice group and that of CD^+_8 cells was significantly increased, thus leading to a definite lowering of the CD_4/CD_8 ratio. It suggests that Cyclosporin A shows immunoregulatory effect on the T.spiralis infection in mice.
