益心饮保护离体大鼠心肌缺血再灌注损伤的研究 被引量:1

摘要 目的采用大鼠离体工作心脏,观察不同浓度的中药复方益心饮对大鼠心肌缺血再灌注损伤的保护作用。方法将36只Wistar大鼠随机分为3组,益心饮I组(平衡液中含0.01g·ml-1中药生药),益心饮II组(平衡液中含0.1g·ml-1中药生药)和对照组;以左室内压峰值(LVSP),左室内压最大上升和下降速率(±dp/dtmax),左室舒张末期压力(LVEDP),心率(HR),心肌组织的丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性的变化作为观察指标并行光镜、电镜观察组织形态学改变。结果实验显示益心饮可明显改善心肌缺血再灌注后的心肌力学和血液动力学等指标,并可降低心肌组织的MDA以及SOD活性含量,减少氧自由基的产生,减轻再灌注的损伤,益心饮II组作用明显优于对照组,P<0.05,心肌组织结构及Ca2+分布异常较其他各组明显减轻。结论中药复方益心饮具有保护心肌缺血再灌注的损伤作用,且其保护作用随浓度的增加而增强。 Aim This study aims to observe the protective effect of Yixinyin on myocardial ischemia-reperfusion injuries in isolated working hearts of tats.Method 36 rats were randomly divided into three groups:Yixinyin group 1 (K-H fluid contain 0.01g·ml^(-1) Yixinyin ),Yixinyin group 2 (K-H fluid contain 0.1g·ml^(-1) Yixinyin) and control group.Myocardial founction was investigated by left ventricular systolic pressure (LVSP),rising and descending rate of left ventricular pressure (±dp/dt(max)),left ventricular end diastolic pressure (LVEDP),heart rate;tissue levels of malondialdehyde(MDA)as well as activity of super oxide demitasse(SOD) were measured in order to evaluate the changes of oxygen free radical metabolism; electron microscope and light microscope were performed to investigate the impairment of myocardial structure and Ca^(2+) distribution.Results Myocardial dynamics and homodynamic indexes LVSP and ±dp/dt(max) were improved siganificantly in Yixinyin group2;the content of MDA in Yixinyin group 2 was obviously lower than that in control group(P<0.05). Yixinyin reduced abnormal of Myocardial structure and Ca^(2+) distribution by comparison with other groups.Conclusion Yixinyin have protective effects against the injury of myocardial ischemia-reperfusion.The effects increase when concentration of Yixinyin increased.
