红车轴草异黄酮对去势大鼠骨质疏松影响的实验研究 被引量:13

摘要 目的:观察红车轴草异黄酮对绝经后骨质疏松症(PMO)的影响。方法:将雌性大鼠去势复制成PMO动物模型,并随机分为6组:即大、中、小剂量红车轴草异黄酮组、阴性对照组、尼尔雌醇组和假手术组。灌胃给药后,用放射免疫法测定大鼠血清雌三醇(E3)、雌二醇(E2)及骨钙素(BGP)水平,用定量超声法测定股骨超声振幅衰减(BUA),并测定大鼠股骨重量以计算股骨干重指数及灰重/干重。结果:红车轴草异黄酮能使去势大鼠血清E3水平升高,血清BGP水平降低,并提高股骨重量和超声振幅衰减。结论:红车轴草异黄酮通过提高血清雌激素水平,增加成骨细胞活性,降低骨高转换率以及减少骨量丢失等多种途径,能有效防治绝经后骨质疏松症。 Objective: To observe the effect of isoflavones in red clover on postmenopausal osteoporosis. Method: The model of postmenopausal osteoporosis was established with ovariectomized (OVX) female Wistar rats. The rats were divided randomly into 6 groups: isoflavones groups with three different doses (L-ISO, M-ISO, H-ISO) and Nilestriol group, ovariectomized group, sham-operated group (SHAM) as control. After drug-given, the serum sex hormone (E3, E2) and osteocalcin (BGP) levels were detected by radioimmunoassay, and the broadband ultrasound attenuation (BUA) of femur was tested with quantitative ultrasound (QUS) , the femur weight was calculated. Result: Isoflavones in red clover could improve level of serum E3 in OVX rats, decrease the content of BGP in serum. Also, the weight and BUA of femur were rised in OVX rats. Conclusion: By means of increasing serum estrogen contents and the activities of osteoblast (OB) , reducing the high turnover rate of bone and preventing the bone mass loss isoflavones in red clover have a effect to prevent and treat postmenopausal osteoporosis.
