摘要
目的 探讨血液学分析中测量不确定度在红细胞计数方面的应用。方法 利用国际临床血液学标准化委员会(ICSH)推荐的参考方法计数血液中的红细胞,并利用相关实验数据和文献资料,按照国标标准化组织(ISO)《测量不确定度表述指南》文件中有关测量不确定度的评估方法,并参考EURACHEM/CITAC《分析测量中不确定度的量化》文件中关于测量中不确定度的评估要求,计算红细胞计数过程中的不确定度。结果 通过对红细胞计数参考方法的不确定度评估,校准物和质控物红细胞计数的扩展不确定度和95%可信区间分别为(4. 26±0. 09)×1012 /L和(4. 27±0. 09)×1012 /L(扩展因子k=2)。结论 本实验评价了影响参考方法红细胞计数的各个分量的不确定度,所占比例最大因素为颗粒计数仪的校准,其次为计数仪计数量和稀释红细胞量。通过对校准物和质控物中红细胞计数值的不确定度进行比较研究,发现在规定了测量过程和分析样品类型时,不确定度能适用于所有测量值,能够保证实验室间结果的可比性。
Objective To study the uncertainty of erythrocyte counts by reference method. Methods By the reference method for enumeration of erythrocyte recommended by ICSH, and some other related experimental and documentary data as well, and according to ISO guide to Expression of Uncertainty in measure, and EURACHEM/CITAC guide quantifying uncertainty in analytical measurement, the uncertainty was analysed in erythroyte counts. Results Afer evaluating the uncertainty the expansionary uncertainty and 95% reliability interval for calibrator was ((4.26)±(0.09))×10^(12)/L (expand factor k=2), for quality control material was ((4.27)±(0.09))×10^(12)/L(expand factor k=2). Conclusions Our study shows that the Z2 particle counter is the most important factor which affects the uncertainty then is the count volume and erythrocytes dluted volume. It also shows that uncertainty is applicable to all the measure values if analysis process and analysis sample are limited.
