Effects of activated ERK on the cultured vascular smooth muscle cells and mechanism

摘要 目的观察磷酸化细胞外信号调节激酶(ERK)对离体培养的平滑肌细胞增殖、凋亡和表型改变的影响。方法培养人大隐静脉平滑肌细胞,实验组培养液中加入PD98059对磷酸化ERK进行阻断后,观测平滑肌细胞的增殖活性,凋亡细胞所占比例以及细胞表型,并与对照组比较。结果实验组与对照组相比,细胞增殖活性明显低于对照组(P<0.01),电镜和荧光染色测定凋亡细胞所占比例明显高于对照组(P<0.01),电镜检测合成型细胞所占比例明显低于对照组(P<0.01),收缩型细胞所占比例较对照组明显为高(P<0.01),α肌动蛋白免疫荧光化学检测显示处理组荧光表达强于对照组。结论对离体培养的人大隐静脉平滑肌细胞磷酸化ERK进行阻断后,平滑肌细胞的增殖受抑,凋亡增加,细胞由合成型向收缩型转变。 Objective To investigate the role of activated ERK on the proliferation,apoptosis and phenotype of the cultured vascular smooth muscle cells.Methods The smooth muscle cells were isolated from the great saphenous vein.The activated ERK was inhibited with PD98059.The cellular proliferation,apoptosis and phenotype were observed.Results Compared with the control group,the proliferative ability of the smooth muscle cells was decreased (P< 0.01),the rate of apoptosis was increased (P< 0.01),the percentage of the contractile cells was increased and the percentage of synthetic cells decreased (P< 0.01),the expression level of α-actin was increased in the experimental group.Conclusion After the activated ERK was inhibited,the degree of cellular proliferation was decreased,the degree of apoptosis increased and the phenotype changed from synthetic to contractile.
