摘要
目的:建立高效液相色谱法(HPLC)测定新疆软紫草中咖啡酸四聚体含量的方法。方法:采用YMCODSAC18色谱柱(4.6mm×250mm,10μm),流动相:甲醇∶水∶甲酸(29∶71∶0.1),流速:1.0ml/min,检测波长:252nm,柱温:40℃。结果:咖啡酸四聚体进样量在0.5~3.5μg范围内与峰面积线性关系良好,相关系数r=0.9995。平均回收率为101.8%,RSD为2.46%(n=9)。结论:高效液相色谱法适用于新疆软紫草中咖啡酸四聚体含量的测定。
Objective: To establish high-perfermance liguid chromatography (HPLC) method of determing caffeic acid tetramer in Arnebia euchroma(Royle) Johnston. Methods: YMC ODS-A C-18 column(4.6×250 mm,10 μm)was eluted using MeOH-H\-2O-HCO\-2H(29∶71∶0.1)as the mobile phase at the column temperature 40℃. The flow rate was 1ml/min and the determination wavelength was 252 nm. Results: The method was proved to be linear in the range from 0.5~3.5 μg with a correlation coefficient of 0.999 5 . The average recovery was 101.8% with RSD of 2.46%(n=9). Conclusion: The method can be applied to the determination of caffeic acid tetramer in Arnebia euchroma(Royle) Johnston..
