小鼠创伤后感染海洋细菌实验模型的建立 被引量:4

Establishment of an experimental model to evaluate the sea bacillus’infectious ability to wounded mouses

摘要 目的:建立小鼠腿部创伤后海水浸泡实验模型并检测海洋细菌对其感染能力。方法:45只CD-1(ICR)VAF小鼠,随机分为实验组(浸泡于含有海洋细菌的人工海水)和阴性对照组(浸泡于不含微生物的人工海水)。致伤后浸泡45min,放入动物洁净饲养柜中饲养,观察浸泡后6h、12h、24h、48h、72h的生命指标及白细胞计数、血培养、伤口分泌物培养。3d活杀或死亡后立即取心,肝,肺等脏器压印培养,病理检查及核酸指纹图分析。结果:实验组致伤浸泡后小鼠精神萎靡,呼吸急促,弓背竖毛,但症状轻重及持续时间因菌种不同而有差异。12h内两组白细胞升高,实验组明显,24h后两组均渐恢复,48h后缓慢接近伤前水平。实验组伤口分泌物培养阳性率100%,副溶血弧菌组12h血培养阳性。实验组小鼠心,肝,肺压印培养分离出的细菌,经DNA指纹图分析证实与相应浸泡菌一致。病理学检查实验组皮下及横纹肌内大量嗜中性粒细胞浸润,肌纤维破坏,蜂窝组织炎形成,对照组少量中性粒细胞散在浸润,横纹肌组织正常。结论:该模型对探讨海洋细菌致病性具有良好的应用前景。 Objective:To establish a wound-and-dip model to evaluate the infectious ability of sea bacillus to wounded mouses. Methods:45 wounded SPF mouses were randomly divided into two groups: the experiment groups(dipped in manual seawater with different sea bacillus) and the control group(diped in the manual seawater without microbe). After being wounded, mouses were diped for 45 minutes, then feeded in clean feeding cages. 6、12、24、48h and 72h after dipping, life indexes、leucocyte counting、blood cultivation、wound secretion cultivation were observed. Being dead or killed three days later, their hearts,livers,lungs were taken out immediately to perform viscera bacteria culture、pathological examination and nucleic aicd finger mark analysis. Results:The experiment groups were disprited, short-breathed, back-arching and fur-erecting with certain difference among different bacillus. Leucocyte counting rised in both groups in the first 12 hours while experiment groups were more obvious. leucocyte counting began to recover 24 hours later and nearly reached before-wounded level 48 hours later. Wound secretion cultivation in experiment groups had a 100% positive rate while blood cultivation had a positive result in Vibro.parahaemolyticus group at 12 hours only. Bacillus separated from viscera cultured organs of experiment groups were the same kind with the dipping bacillus,which was confirmed by DNA finger mark analysis. Subcutaneous and the striped muscle of experiment groups were soaked by numerous neutrophiles and muscle fiber destoried, alveolate tissue inflammation formed. In control group, there were only few neutrophile in subcutaneous and the striped muscle with muscle fiber normal. Conclusions:This model had a promising value in the inffectious study of sea bacillus.
