摘要
目的采用高效液相色谱法测定人血浆中甘草次酸浓度。方法建立测定人体血浆中甘草次酸的高效液相色谱法,采用Nova-PakC18柱(150mm×3.9mm,4μm),流动相:2%醋酸-乙腈(45∶55),流速:1mL/min,检测波长:254nm。采静脉血,测定不同时间的血浆药物浓度。结果甘草次酸血药浓度在53~1060ng/mL范围内线性良好,相关系数为0.9995,测得平均回收率为106.67%,日内、日间RSD均小于7%。结论本法适用于人体内甘草次酸的含量测定及药代动力学研究。
Objective To determine the plasma concentration of glycyrrhetic acid (GA) by HPLC. Method To establish an assay for the quantification of GA in human plasma by HPLC with a Nova-Pak C18 column (150 mm×3.9 mm, 4.6 μm) with UV detection at 254 nm, and the flow rate was 1.0 mL/min. Results The calibration curve was linear (r=0.9995) within the range of 53~1060 ng/mL for GA in plasma. The average recovery was 106.67%. The within-day and between-day derivation (RSD) were less than 7%. Conclusion The method can be used to determine the concentration and to investigate the pharmacokinetics of GA in human.
