摘要
本实验在经血清调理酵母聚糖激活的大鼠肺泡巨噬细胞(AM)体外培养4h后,检测其上清液中的PGs含量。消炎痛预处理使激活的AM的释放的PGE1、E2、F2a量显著减少,E2/E1和E2/F2a比值降低;而细胞松弛素B预处理AM则可使上述PGs的含量显著增加。以中性粒细胞趋化指数来反映IL-8的活性,表明消炎痛预处理AM组的IL-8的分泌量显著减少,而细胞松弛素B预处理AM组的IL-8分泌量显著增多。AM的PGs释放量与其IL-8分泌量呈平行的变化关系,说明内源性PGs在AM的分泌IL-8的活动中起着调控作用。
To test the hypothesis that endogenous prostaglandins(PGs) play a role in
modulatingalveolar macrophage-derived interleukin 8(IL-8) secretion, the level of PGs and IL-8
producedby zymosan-activated alveolar macrophages(AM) were exami ned. Groups of AM
pretreatedfor 30 min with indomethacin(10μg/ml) or cytochalasin B(4μg/ml) were incubated for
4h withRPMI-1640 containing serum opsonized zymosan (OPZ, 200μg/ml), Supematant were
thenremoved to quantitate PGE1,E2,F2a and TXB2 by RIA and to measure the chemotactic
activi- ties of IL-8 to neutrophils(purified from human peripheral blood) by the modified Boyden
tech-nique using blindwell chemotaxis chambers. It was found that the contents of PGE1,E2,F2a
andratio of PGE2/E1 and E2/F2a in indomethacin pretreated group were decreased, and
contentsof PGE1,E2, F2a were increased in cytochalasin B pretreated group.Changes of
neutrophilchemotactic index to IL-8 in incubated AM supernatants showed the same
direction,inhibitionin indomethacin pretreated group and enhaneement in cytochalasin B
pretreated group. The re-sults show that there is a close relationship between activity of IL-8
and content of PGs producedby AM. It is concluded that endogenous PGs may play an important
role in secretion of AM-de-rived IL-8 in host defense and disease.
