小鼠大脑皮质N─甲基-D-天冬氨酸受体的鉴定及其在衰老时的变化 被引量:4

0IDENTIFICATION OF NMDA RECEPTOR IN CEREBRAL CORTEX OF MICE AND ITS CHANGE DURING AGING

摘要 以放射性配基结合分析法对正常成年小鼠大脑皮质中N─甲基-D-天冬氨酸(NMDA)受体作了鉴定;观察了衰老小鼠NMDA受体、空间辨别能力、海马突触传递长时程增强(longtermpotentiation,LTP)的变化及补肾中药复方对这些变化的影响。结果表明:小鼠大脑皮质含有丰富的、高亲和力的NMDA受体;衰老过程中小鼠NMDA受体的最大结合容量(Bmax)呈渐进性降低,空间辨别能力下降,LTP的振幅和斜率明显降低;补肾中药复方具有提高衰老小鼠NMDA全体Bmax值和维持LTP于较高水平的作用。 By means of radioligand binding assay,the N-methyl-D-aspartate(NMDA) receptor wasidentified in the cerebral cortex of mature mice, and the change of NMDA receptors in aged micewas observed.Meanwhile the influence of kidney-tonifying apozem consisting of eight kinds ofChinese medicinal herbs on this change was investigated. The results demonstrated that abundantNMDA receptors with high affinity existed in the cerebral cortex of mice. In aged mice themaximal binding capacity(Bmax)of NMDA recptors and the amplitude and slope of long termpotentiation(LTP)decreased markedly. Administration of kidney-tonifying apozem could im-prove the changes of NMDA recptors and LTP in aged mice. Upon analyzing the results ob-tained,the following conclusions can be reached:(1) The change of NMDA receptors may be re-lated with the decline of learning and memory ability during aging.(2) Administration ofkidney-tonifying apozem can improve the ability of leaming and memory during aging.
