羊膜移植对实验性单疱病毒角膜炎中MMP-9及TIMP-1表达的影响 被引量:4

Expression of matrix metal loproteinase-9 and tissue inhibitor of metalloproteinase-1 in her-petic stromal keratitis mouse model with am-niotic membrane transplantation

摘要 目的探讨羊膜移植对单疱病毒性角膜炎中基质金属蛋白酶(MMP-9)及其抑制剂(TIMP-1)在小鼠角膜中的表达影响及意义。方法将40只Balb/c小鼠随机分为实验组和对照组,将5μL含有100×103pfuHSV-1病毒的DMEM滴于2组小鼠右眼划伤角膜表面建立单疱病毒性角膜炎模型,对实验组小鼠右眼角膜行羊膜移植术;对照组不行羊膜移植。在羊膜移植术后0、2d、7d、14d处死动物,用免疫组织化学染色方法和计算机图像分析系统检测2组小鼠MMP-9及TIMP-1在角膜中的分布及其平均光度值。结果羊膜移植组临床评分显示各时间点角膜上皮病变、角膜混浊和新生血管明显低于对照组。免疫组织化学染色对照组角膜中MMP-9在第2d出现表达,第14d达高峰。TIMl-1开始表达不明显,第7d出现表达,14d达高峰。羊膜移植组各时间点MMP-9表达低于对照组,而TIMP-1表达高于对照组。结论羊膜移植术可通过调节基质金属蛋白酶及其抑制剂的表达平衡,从而抑制和延迟HSV感染后单纯疱疹病毒性角膜炎的发生和发展。 Objective To study the effects of amniotic membrane transplantation on the expressions of matrix metalloproteinase-9 (MMP-9) and its tissue inhibitor(TIMP-1) in mice with herpetic stromal keratitis(HSK). Methods Forty Balb/c mice were randomly divided into two groups and inflected with HSV-1. Amniotic membrane transplantation(AMT)was performed in AMT group.At different time (0,2 days,7 days,14 days) after AMT,the expressions of MMP-9 and TIMP-1 in corneas were detected by immunohistochemistry ,and the results were analyzed by computer medical image analysis system. Results In control group, the level of MMP-9 elevated on the 2nd day, peak on the 14th day.TIMP-1 was expressed on the 7th day,reached peak on the 14th day. At the same time points, the levels of MMP-9 in AMT group were greatly lower than those in the control group, while the levels of TIMP-1 higher than those in the control group. Conclusion Immediate intervention for HSK with amniotic membrane transplantation could promote wound healing and inhibit the development of cornea melting by adjusting the proportion of matrix metallo-proteinase and its tissue inhibitor.
