实验性单纯疱疹性角膜炎中基质金属蛋白酶mRNA的表达

摘要 目的 :探讨Ⅰ型单纯疱疹病毒 (HSV 1)所致的角膜炎中基质金属蛋白酶 (MMPs)mRNA的表达。方法 :雌性BalB/c小鼠眼角膜接种HSV 1(KOS株 )以诱发单纯疱疹性角膜炎 (HSK)。收集正常角膜及感染后第 2 ,7,14d的感染角膜 ,应用RT PCR技术检测基质金属蛋白酶 2 ,基质金属蛋白酶 9(MMP 2 ,MMP 9)的mRNA表达。结果 :除正常角膜MMP 9mRNA为阴性表达其他各组均获得阳性目的片断。感染后第 2dMMP 2mRNA表达较正常组差异没有显著性 (P >0 .0 5 ) ,感染后第 7dMMP 9mRNA表达与第 2d的表达差异没有显著性 (P >0 .0 5 ) ,其他各组差异均有显著性意义 (P <0 .0 1)且大体上呈随感染天数增加表达增高的趋势。结论 :在实验性HSK中存在基质金属蛋白酶mRNA的表达且MMP 2 ,MMP \|9的mRNA高表达参与了角膜溃疡、角膜新生血管的形成等病变的发生。 Objective: To explore the expression of matrix metalloproteinases (MMPs) mRNA in corneas with infection of herpes simplex virus type-1(HSV-1). Methods: Keratitis was induced in BalB/c mice by inoculating the cornea with HSV-1(KOS strain).Corneas were harvested before infection and at day 2,7 and 14 post-infection. The expression of MMP-2 and MMP-9 mRNA were examined by reverse transcription-polymerase chain reaction (RT-PCR). Results: Each groups showed positive segments except the expression of MMP\|9 mRNA in the normal group. The MMP-2 mRNA at day 2 post-infection did not express significantly compared with the normal group( P> 0.05).The MMP-9 mRNA at day 7 post-infection did not express significantly compared with the 2 day post-infection group( P> 0.05).The differences among other groups were statistically significant ( P< 0.01) and they presented the trend that the expression increased with the time passed post-infection on the whole. Conclusion: The expression of the MMP-2 and MMP-9 mRNA can be detected in HSK cornea. This enzyme was shown to contribute to the ulcerative and neovascularization process that occurs in the corneal in response to HSV infection.
