白细胞介素6对甲状腺细胞钠/碘转运体的基因调控 被引量:1

摘要 目的研究白细胞介素6(IL-6)对甲状腺滤泡细胞膜上钠/碘转运体(NIS)基因表达的影响,探讨细胞因子在甲状腺生理功能调控和甲状腺疾病发病机制中的作用和地位。方法取Graves病(GD)患者的甲状腺组织,进行甲状腺细胞原代培养,培养5d后,吸去培养液,换用含不同浓度IL6的试验液,与单层培养的滤泡上皮细胞共同孵育48h,运用半定量逆转录聚合酶链反应(RTPCR)技术检测NISmRNA表达的变化。结果IL-6浓度在0U/ml、1U/ml时,GD甲状腺滤泡上皮细胞膜上NISmRNA表达无明显差异;IL-6浓度在10U/ml、100U/ml时,NIS基因表达比前一浓度时明显受抑制(P<0.05),且高浓度比低浓度抑制更加明显;IL-6浓度为1000U/ml时,NIS基因表达量与前一浓度比较无统计学差别。结论IL6可抑制甲状腺滤泡细胞基底膜上NIS基因的表达,提示细胞因子IL-6的浓度不同在甲状腺生理功能调控中发挥的作用也不同。 Objective To investigate the influence of interleukine-6 (IL-6) on sodium iodide symporter (NIS) mRNA expression in the thyroid cells from the patients with Graves' disease (GD). Methods Thyrocytes from GD were cultured in absence or presence of different concentrations of IL-6 for 48h. The level of NIS gene expression was assessed by reverse transcription polymerase chain reaction(RT-PCR) method.Results IL-6 inhibited NIS mRNA in the thyroid cells in a dose- dependent manner.Conclusion IL-6 plays an important role in regulation of thyroid function and thyroid homeostasis.
