白细胞介素-1β诱导牛脑微血管平滑肌细胞增殖及药物的拮抗作用 被引量:2

摘要 研究白细胞介素-1β(IL-1β)对牛脑微血管平滑肌细胞(BCSMC)增殖的影响。浓度为5~100ng·L-1的IL-1β与BCSMC共同孵育24h,即可明显诱导BCSMC的增殖,IL-1β浓度为50ng·L-1时,增殖率为17%。欧芹素乙,异欧芹素乙,6-(α,α-二苯基乙酰哌嗪基苯基-4,5-二氢-5-甲基-3(2H)-哒嗪酮(简称DMDP),6-(α-本基乙酰哌嗪基苯基)-4,5-二氢-5-甲基-3(2H)-哒嗪酮(简称PMDP)分别在不同浓度可显著抑制IL-1β所诱导的BCSMC增殖。 The effects of Interleukin-1beta(IL-1β) on the proliferation of bovine cerebromicrovscular smooth cells (BCS MC) were investigated. After 24 h incuba tion with BCSMC IL-1β induced prolifera tion of cultured BCSMC at concentration from 5 to 100 ng· L-1. At 50 ng·L-1,after 24 h incubation with BCSMC, stimulative effect of IL-1β reached 17%. It is found that imperatorin(Imp), isoimperatorin(Isi).6-(αα-diphenylacetylpipezinyl)phenyl-5-methy-4,5-dihydro-3(2H)-pyridazinone (DMDP)and 6-(α-phylly lacetylpipezinyl)phenyl-5-methy-4,5-dihydro-3(2H)-pyridazinone (PMDP0respectively antagonized IL-1β induced prolifera tion of BCSMC at different concentrations.At 10-6 mol·L-1, the inhibitory rated ofImp, DMDP and PMDP were respectively16. 5%,22.44% and 24.09%;At 10-5 mol·L-1, the inhibitory rates of Imp, Isi andDMDP were respectively 38.28%,17,49%and 13.20%.At 10-4 mol· L-1, the inhibitory rates of Imp,Isi and Ver were respectively 30.69%,20. 79% and 31.68%.
