摘要
目的 研究组织工程皮肤培养液中IL 1β、IL 6和IL 8的分泌情况。方法 收集组织工程皮肤制备过程中单层成纤维细胞(fibroblast ,FB)培养液、胶原凝胶真皮替代物(dermalequivalent,DE)、复合壳多糖DE和复合壳多糖皮肤替代物(skinequivalent,SE)培养液标本,ELISA法检测其中IL 1β、IL 6和IL 8分泌量。结果 复合壳多糖真皮基质促进FB、角质形成细胞(keratinocyteKC)分泌IL 6、IL 8。SE分泌的IL 6、IL 8在DE加KC之后和SE形成分层表皮时形成两个高峰。结论 FB、KC在壳多糖组织工程皮肤中混合器官型培养时分泌IL 6和IL 8较明显,这些细胞因子可在创面愈合中发挥重要作用。
Objective To investigate the secretion of IL-1β, IL-6 and IL-8 in cultured tissue-engineered skin. Methods IL-1β, IL-6 and IL-8 from the culture medium of monolayer fibroblast (FB), collagen dermal equivalent (DE), composite chitosan DE, and composite chitosan skin equivalent (SE) were detected by ELISA. Results Composite chitosan dermal matrix could promote FB and keratinocyte (KC) to secrete IL-6, IL-8. There were two rush hours of IL-6, IL-8 secretion from SE, after KC to DE and epidermal stratification of SE were added. Conclusion Chitosan can facilitate FB and KC to secrete IL-6 and IL-8 in cultured tissue engineering skin and benefit wound healing process.
