Enantiomeric Separation of Trimebutine Maleate by High Performance Liquid Chromatography Using α_1-Acid Glycoprotein Chiral Stationary Phase

摘要 以α1AGP为固定相,建立了马来酸曲美布汀的HPLC对映体拆分方法。考察了流动相pH、缓冲盐溶液的浓度、有机改性剂种类和浓度、流速、柱温等对对映体分离的影响。优化后的色谱条件为:ChiralAGP(100mm×4.0mm,5μm)色谱柱,流动相为10mmol/L醋酸铵缓冲液(pH3.8);流速为0.4mL/min;柱温为35℃。马来酸曲美布汀对映体的保留时间分别为7.4min和9.8min;分离度为1.59。 The enantiomeric separation of trimebutine maleate(TBM) was investigated by high performance liquid chromatography (HPLC) on a α-AGP chiral stationary phase. The influence of the pH value and flow rate of the mobile phase, the concentration of buffer solution and organic solvents and temperature were examined . The best separation was obtained with 10 mmol/L ammonium acetate (pH 3.8) buffer as mobile phase with the flow rate set at 0.4 mL/min and the temperature of 35℃. The retention time of the two TBM enantiomers was 7.4 min and 9.8 min, respectively,a resolution of 1.59 was achieved.
