摘要
目的制备莪术油纳米乳剂,详细评价制备工艺中影响莪术油纳米乳剂质量的因素,并考察纳米乳剂在不同释放介质中的药物释放。方法采用高压乳匀法制备莪术油纳米乳剂;采用单因素和正交设计法考察制备工艺中影响莪术油纳米乳剂质量的因素;采用动力光散射法测定其粒径及粒径分布;采用透射电镜法观察其形态;采用葡聚糖凝胶柱层析法测定包封率。结果所制备的纳米乳剂为类球状,大小均匀。平均粒径为( 76 5±10 1)nm ,纳米乳剂中吉马酮的质量浓度为( 1 76±0 0 9)g·L-1,吉马酮的包封率为( 84 4±1 0 ) %。结论高压乳匀法可用于莪术油纳米乳剂的制备。
Objective To prepare curcuma zedoaria oil-loaded nanoemulsion and investigate its physicochemical properties.Methods The high pressure homogenizer method was employed to prepared Curcuma zedoaria oil-loaded nanoemulsion by single factor exploration and orthogonal design.The particle size distribution was investigated by dynamic light scattering(DLS) method.The sephadex gel chromatography method was established for determination of drug entrapment efficiency (EE) of germacrone in the nanoemulsion.The morphological investigations of nanoemulsion were performed by transmission electron microscopy (TEM).Results Curcuma zedoaria oil-loaded nanoemulsion prepared by high pressure homogenizer method was spherelike with the mean particle size of (76.5±10.1)nm.The content of germacrone in the nanoemulsion was (1.76±0.09)g·L~ -1 ,drug entrapment efficiency was (84.4±1.0)%.Conclusions The high pressure homoenizer method is available for the preparation of Curcuma zedoaria oil-loaded nanoemulsion,a satisfied effect can be achieved by adjusting the formulation and process parameters.
