摘要
目的研究2型糖尿病大鼠肾组织骨形态发生蛋白-7(BMP-7)和转化生长因子-β1(TGF-β1)表达的关系。方法将大鼠模型28只随机分为正常对照组、糖尿病组、治疗组。用RT—PCR方法研究肾脏组织中BMP-7和TGF-β1 mRNA水平的变化及其关系,用SPSS 11.5软件进行医学统计和相关分析。结果采用产物和内参(β-actin)的吸光度容积比值作为表达强度。糖尿病组大鼠BMP-7表达较正常组明显降低,TGF-B1表达明显升高,二者呈显著负相关(r=-0.819 P<0 01)。结论BMP-7和TGF-β1在糖尿病大鼠肾组织中表达变化相关,二者相互作用调节肾脏内环境平衡,在糖尿病肾病发生、发展中起重要作用。实用儿科临床杂志,2005,20(5)
Objective To investigate the relationship between the expression of bone morphogenetic protein-7(BMP-7) and the expression of transform growth factor-β1 (TGF-β1) in kidney tissue of type 2 diabetes rats Methods The rats were devided into control group,diabetes mellitus group and treated group. The expression of BMP-7 and TGF-β1 in kidney tissue and the relationship between them were studied by reverse transcript(RT) - PCR method. The medical statistics and correlation analysis were done by using SPSS 11.5 analysis software. Results The ratio of the optical density volume of the produce and inner reference (β-actin) was used as the level of expression. The level of expression of BMP-7 in the diabetes mellitus group was less than it in normal group markedly, but the level of expression of TGF-β1 was just reverse. The expression of the BMP-7 and TGF-β1 also took on negative correlation significantly (r= -0.819 P<0.01).Conclusions The change of the expression of BMP-7 in kidney tissue of diabetes rats is accorded with that of TGF-β1 and the two factors interacted and regulated the balance of inner environment in kidney. They take an important role in the occurrence and progression of diabetic nephropathy. J Appl Clin Pediatr, 2005,20(5): 397 - 399
