白蛋白对内皮细胞单层屏障功能的增强作用 被引量:5

摘要 我们建立了体外灌流内皮细胞单层研究血管通透性的方法,可以测定内皮单层的滤过系数(Kf)和渗透压反射系数(σ),研究了白蛋白对内皮单层通透屏障功能的影响。不同浓度白蛋白溶液(1,5,10,20mg/ml)灌流单纯的滤膜时,其流量变化不大,灌流滤膜上致密生长的内皮细胞单层时,流量和Kf值随白蛋白浓度的增加而降低。同时,蛋白清除率也不断减小,σ增大,说明白蛋白通过与内皮细胞的作用使内皮单层对水和大分子物质的通透性降低,对内皮通透屏障功能有增强作用。 A method was established to study vascular permeability on pulmonaryartery endothelial cell monolayers perfused in vitro.Filtration coefficient(Kf) and osmoticreflection coefficient(ORC)were determined and the effects of albumin on the permeabilitybarrier function of endothelial monolayers were studied.When albumin at different con-centrations(1,5,10 and 20mg/ml respectively)in Hanks'balanced salt solution(HBSS)were used to perfuse gelatinized filter membrane alone,the flux volume did not changeobviously.When confluent endothelial monolayers were perfused, flux volume,Kf valueand protein clearance rate decreased and ORC increased markedly as albumin concentrationincreased.These results demonstrated that albumin decreases endothelial monolayer permea-bility to water and macromolecules and suggested that it may enhance microvascular permea-bility barrier function.
