CBA/N、nude和SCID小鼠肝癌模型生长和凋亡基因表达及其相关性

The Development of Hepatocarcinoma Model and Expression of Apoptotic Gene and Their Relationship in CBA/N,Nude and SCID Mice

摘要 目的研究CBA/N、nude和SCID小鼠人肝癌模型的特点,凋亡相关基因的表达及其相关关系。方法用人肝癌细胞系HHCC9724P接种于动物肝脏,于各时间点观察肿瘤体积的生长情况。用免疫组织ABC法对其bcl2,bax和VEGF的表达及其与肿瘤组织体积相关性进行研究分析。结果3种动物肝内种植肝癌细胞后均可成活,bcl2,bax和VEGF的表达明显增强(P<0.01)。肝癌细胞种植3周后肿瘤体积和基因表达达最高水平。其中SCID小鼠尤为突出(P<0.01)。3种小鼠肝内肿瘤体积发展与3种基因表达改变相关性分析结果证明具有明显的正相关关系。结论SCID小鼠的人肝癌细胞种植成功率和凋亡基因表达最为显著。肝内肿瘤的生长与凋亡基因表达具有明显的正相关关系。 Objective To study the models of hepatocarcinoma and the expression of apoptotic gene and their relationship in CBA/A,nude and SCID mice.Methods The hepatocarcioma cells (HHCC-9724-P) were inoculated to the liver of CBA/N,nude and SCID mice,the size of the tumor tissue was measured in different times and the expression of bcl-2,VEGF and bax were observed by means of immunohistochemical ABC method and its relationship to the tumor’s size and expression of apoptopic genes also were studied.Results The human hepatocarcinoma cells could inoculated successfully in the liver of CBA/N,nude and SCID mice. The size of the tumor tissue and the expression of bcl-2,bax and VEG increased and reached a peak for 3 weeks but the SCID mice was the most obvious.The positive cells rate bcl-2 and VEGF was higher than that of bax.The development of tumor size closely related to the expression of bcl-2,bax and VEGF.Conclusion SCID mice is the best animal in the human hepatocarcinoma cell inoculation successful rate and the expression of apoptotic gene.The development of the tumor tissue is closely related to the expression of apoptotic gene.
