摘要
目的 通过比较氧苯达唑(Oxibendazole, OBZ)与阿苯达唑(Albendazole,ABZ)灌注治疗小白鼠细粒棘球蚴病(Echinococcosis),观察OBZ抗小白鼠体内细粒棘球蚴的药效。 方法 采用随机对照试验方法,将感染细粒棘球蚴 10周的78只小白鼠随机分为5个组:1)阿苯达唑粉剂组(Po ABZ)75.0 mg/kg;2) OBZ 37.5 mg/kg 组;3)OBZ 75.0 mg/kg 组;4)OBZ 150.0 mg/kg 组;5) 生理盐水对照组(NS),每次0.3 ml/只。每周用药3次,连续用药10周后剖检。测定棘球蚴囊湿重和减重率;观察棘球蚴囊的病理形态学改变及超微结构变化,用 HPLC法测定血液、肝脏、肺脏中的药物浓度。 结果 OBZ低剂量(37.5 mg/kg)、中剂量(75.0 mg/kg)和高剂量(150.0 mg/kg)组小鼠棘球蚴囊平均湿重分别为(159.33±263. 21) mg、(49. 75±43. 62) mg 和(191. 5±98. 36) mg,平均减重率分别为 82. 49%、94. 53%和78.96%。ABZ组平均囊湿重和减重率分别为(145.13±162.47) mg和84.05%;各药物治疗组与 NS对照组(910.11±1 496.28) mg比较差异均有显著性(P<0.05);OBZ 75.0 mg/kg、150.0 mg/kg组与ABZ(75.0 mg/kg)比较差异无显著性(P>0.05)。OBZ不同剂量组小鼠囊壁病理改变只数和程度与空白对照组比较差异均有显著性,而与 ABZ组比较差异无显著性。 结论 OBZ具有?
Objective To observe the efficacy of Oxibendazole (OBZ) against secondarily infected Echinococcosis in mice. Methods The 78 mice infected Echinococcus granulosus (E.g) for 10 weeks randomly divided into five groups: 1) Albendazole (ABZ) oral administration with 75 mg/kg dosage; 2)OBZ oral administration with 37.5 mg/kg dosage; 3)OBZ oral administration with 75 mg/kg dosage; 4) OBZ oral administration with 150 mg/kg dosage; 5) Saline oral administration with 0.3 ml per mouse dosage. All groups were administered with 3 times a week medication for 10 weeks. Following investigating markers were used after overall administration: 1) Mean cyst weight and reduction rate after treatment; 2) Histopathological changes of the cyst under both optical and electron microscope; 3) Measurement of the concentrations of the OBZ, ABZ and its metabolites, ABZ sulfone (ABZSN) and ABZ sulfoxide ( ABZSF), in plasma, liver and lung tissues. Results The mean cyst weights and reduction rates were (159.33± 263.21 ) mg and 82.49%, (49.75±43.62) mg and 94.53%, (191.5±98.36) mg and 78.96% in different OBZ dosages with 37.5 mg/kg, 75 mg/kg and 150 mg/kg groups respectively, and (145.13±162.47) mg and 84.05% in ABZ group. There were significant differences of mean cyst weights between drug groups and control group( P <0.05), but not between OBZ and ABZ groups( P > 0.05 ).There were significant difference of histopathological changes of E. grnulosus cyst between different dosages in OBZ groups, ABZ group and saline group ( P <0.05), but there was no difference between the OBZ (75 mg/kg) group and ABZ (75 mg/kg) group ( P >0.05). Conclusion OBZ was highly effective against E. granulosus cyst in mice, the efficacy of OBZ is about to equal with ABZ.
