环维黄杨星D对高血压大鼠脑缺血损伤的保护作用 被引量:11

Protective effect of cyclovirobuxine D(环维黄杨星D) on cerebral ischemic injury induced by hypertension in rats

摘要 目的:观察中药单体环维黄杨星D(CVB- D)对易卒中型肾血管性高血压大鼠(RHRSP)脑缺血损伤的保护作用。方法:采用环形银夹造成SD大鼠的双侧肾动脉狭窄,制成RHRSP;再用线栓法制成一侧大脑中动脉闭塞(MCAO)脑缺血再灌注模型。观察CVB- D治疗对脑缺血6 h后再灌注6、72和16 8h不同时间点大鼠的神经行为、脑梗死面积及脑含水量等影响,并与生理盐水组进行对照。结果:脑缺血再灌注6、72和16 8h后,CVB D治疗组大鼠脑含水量分别为(79.0 8±1.0 4 ) %、(80 .84±1.2 7) %和(78.37±1.16 ) % ,比同期对照组脑含水量(82 .32±1.6 1) %、(83.88±1.4 8) %、(81.6 1±1.31) %明显下降(P<0 .0 5或P<0 .0 1) ;缺血再灌注72及16 8h后治疗组大鼠脑梗死面积分别为(14 .4 1±1.2 6 ) %和(18.4 1±1.6 2 ) % ,明显低于同期对照组脑梗死面积(2 1.4 2±1.6 1) %、(2 2 .77±1.4 8) % (P均<0 .0 5 )。结论:CVB D对实验性脑缺血-再灌注大鼠脑损伤有一定保护作用。 Objective: To observe the protective effect of traditional Chinese medicine cyclovirobuxine D(CVBD,环维黄杨星D) on cerebral ischemic injury induced by renovascularhypertension prone to stroke in rats.Methods: The rat renovascularhypertension model prone to stroke was established by clipping bilateral renal arteries with silver loop clips; then a nylon suture was used to ligate an unilateral middle cerebral artery, its occlusion was achieved and a model of cerebral ischemia/reperfusion was established. The neurological behavior, the volume of water content in the brain, and the size of cerebral infarction area were observed 6 hours after ischemia and 6, 72, 168 hours after reperfusion CVBD was applied in treatment group and normal saline, in the control group. The results were compared between the two groups. Results: In the control group, cerebral water content were(82.32±1.61)%, (83.88±1.48)% and (81.61±1.31)% after 6, 72 and 168 hours of reperfusion respectively. Compared with the control group 〔(79.08±1.04)%, (80.84±1.27)% and (78.37±1.16)%〕, the cerebral water content of CVBD treatment group declined significantly (P<0.05 or P<0.01), and the rates of cerebral infarction area 〔(14.41±1.26)% and (18.41±1.62)%〕 were also smaller in the treatment group than that of the control subgroups 〔(21.42±1.61)% and (22.77±1.48)%〕 after 72 and 168 hours of reperfusion (both P<0.05).Conclusion: CVBD has markedly protective effects on experimetal rats with cerebral injury induced by ischemia/reperfusion.
