摘要 目的:建立狗带血管坐骨神经的动物模型,探讨神经、动脉和静脉三种组织同时冷冻保存的可行性及不同维持温度对冷冻保存神经和血管的影响。方法:采用二步冷冻步骤,选择4种不同的维持温度(-40℃、-50℃、-60℃、-70℃)对狗带血管坐骨神经的动物模型进行冷冻,在液氮中保存21 d,取出标本用37℃水浴复温。观察方法包括光镜下HE常规染色和髓鞘染色,四氮唑兰还原试验。结果:经-60℃维持温度冷冻后再保存于液氮中的动脉、静脉、神经,其血管代谢活性,内皮细胞和神经雪旺细胞的保持较好。结论:狗带血管坐骨神经的动物模型,解剖结构清晰,可操作性较好。神经、动脉和静脉三种组织同时冷冻保存是可行的,-60℃是带血管神经冷冻保存较理想的维持温度。 Objective:To establish animal model of vascularized dog sciatic nerve and study the possibility of the cryopreservation of three kinds of tissues (nerve, arteria and vein) at the same time, while explore the effects of different holding temperature on the cryopreservation of nerves and vessels. Methods:Vascularized dog sciatic nerves were stored in liquid nitrogen 21days by a two-step freezing schedule, in which different holding temperature (-40℃ ,-50℃ ,-60℃ ,-70℃) were employed in four groups, and then took out and rewarmed the specimens at 37℃. The methods observing specimens included HE dyeing, myelin sheath dyeing and tetrazolium test. Results:The vascular metabolic activity, the structure of the vascular endothelial cell and Schwann cell after cryopreservation were well preserved at the holding temperature -60℃. Conclusions:The anatomic structure is clear and acceptable for the animal models of vascularized dog sciatic nerve, while the cryopreservation of three kinds of tissues (nerve, artery and vein) is feasible at the same time. -60℃ is the optimal holding temperature for the cryopreservation of vascularized nerve.
