摘要
目的:研究三叶委陵菜、异叶蛇葡萄、蛇葡萄及细叶卷柏提取物对培养细胞的毒性、抗柯萨奇病毒 B3(CVB3)的作用及其机理。方法:通过观察药物毒性、病毒引起的细胞病变效应、MTT法检测细胞活性,判断药物毒性及药物抗病毒效应。结果:4种药物的半数毒性浓度(TC50)分别为7.2, 8.1, 8.7, 12.4μg·ml-1。三叶委陵菜、异叶蛇葡萄、细叶卷柏可直接杀伤病毒,半数抑制浓度(IC50)分别为0.52, 1.21, 0.53μg·ml-1,治疗指数(TI)分别为14, 6.7, 23;蛇葡萄、细叶卷柏可阻止病毒的吸附,IC50分别为0.56, 0.51μg·ml-1,TI分别为16, 23。药物不能抑制病毒的生物合成。结论:4种药物通过不同机制发挥抗CVB3的作用;但是同时对细胞也有一定的毒性作用,仍需进一步研究有效的抗病毒成份。
Objective: To investigate the toxic effect on cells and the anti-Coxackievirus B3(CVB3) effect of Potentilla freyniana, A. humulifolia var. heterophylla Ampelopsis sinica, and Selaginel lalabordei in vitro. Methods: The toxic and antiviral effect was evaluated by observing toxic effect, cytopathic effect (CPE) and MTT colorimetric assay for vital cell rate. Results: Median toxic concentration(TC_ 50 ) of four extracts were 7.2,8.1,8.7 and 12.4 μg·ml -1 respectively. Potentilla freyniana, Ampelopsis sinica and Selaginella labordei showed the ability of directly-killing-virus and the median inhibitory concentration (IC_ 50 ) were 0.52,1.21 and 0.53 μg·ml -1 respectively. So the treatment index (TI) were relatively figured out as 14, 6.7 and 23. Ampelopsis sinica and Selaginella labordei showed the ability of adsorption-blocking and the IC_ 50 were 0.56 and 0.51 μg·ml -1 , respectively. And the TI was 16 and 23 respectively. The extracts showed no abilities of inhibiting viral biosynthesis. Conclusion: The four plants have the activity of anti-CVB3 through several ways. But they also have certain toxic effects on cells in vitro. The effective elements from these plants need to be further studied.
